Abstract |
BRAF-mutantcolorectalcancersformalargetumoursubsetwithdistinctivemorphologic,geneticandclinicalcharacteristics.Themolecularevolutionofthesecancersishoweverpoorlyunderstood.Wedevelopedaconditional(Cre-orSleepingBeautyactivatable)BrafV600Eknock-inmouselineandfoundthatmutantBrafinitiatesanovelpathwaytointestinalcancer,whichprogressesthroughahyperplasia/adenoma/carcinomasequencemimickingkeygeneticandmorphologiccharacteristicsofthehumancondition.ToovercomethelimitationsinherenttoexomesequencingofthesetumorswedevelopedaPiggyBactransposon-basedcancergenediscovertool-kitinmice,whichenablesunbiasedgeneticscreeninginvivo.Usingthisapproachrelevantpathwaysorpathwaybranchesare“hit”bytransposonsatmultiplelevelsandthetransposonintegrationpatternpredictsthegene’sfunctionasoncogenicortumoursuppressive.First,weaimtoperformPiggyBac... |
